Responsible for the discovery, isolation, purification, and structural
elucidation of biologically active compounds from microbial fermentation
mixtures.
- Used solvent extraction and reversed phase, normal phase, gel
filtration, and counter current chromatography to purify possible
active components from microbial fermentation mixtures on both the
preparative and analytical scales.
- When a compound was purified to an acceptable degree, used IR, NMR,
mass spectroscopy and UV/VIS spectroscopy to elucidate the structure of
the active compound.
- Wrote an internal publication describing usage of IR and UV spectrum
library matches to identify an active fermentation compound as a known
compound.
- Developed and ran a biological assay from its conception to its
completion. The assay was used in place of a more complex, costly
assay. Maintained all assay test results in a computer database.
Provided oral and written reports of assay results to those interested
in using the assay. Assay results were written up in an internal
publication.
- Analytical chemistry equipment used: the Varian star 9020 Analytical
HPLC, the Hewlett-Packard 1090 HPLC, the Perkin Elmer 3600 IR, the
Perkin Elmer 7300 IR, the 500 MHz Bruker NMR, the Hewlett-Packard 8450a
UV/VIS Spectrometer, and the Waters Prep Liquid Chromatography 3000
System.
- Programmed a robot to do extractions, pass fermentations through
resins, etc.
- Maintained and updated IR spectral library by digitizing IR spectra
of novel compounds from the literature into the computer library.
